Stimulating Meniscal Cartilage Regeneration Using Recombinant Growth Factors Invitro

چکیده

INTRODUCTION Meniscal tears are common debilitating injuries that can have dramatic consequences especially for young athletes. Healing of meniscal tears following surgical repair is essentially only feasible for tears within the peripheral, vascular, zone. By comparison, tears that occur within the inner, avascular, zone do not heal and most commonly are treated by excision. This in turn greatly increases the potential for secondary osteoarthrosis. Clearly, there is a clinical need to augment surgical techniques, extending meniscal repair to the avascular zone.

Regional regenerative potential of meniscal cartilage exposed to recombinant insulin-like growth factor-I in vitro.

It is well recognised that meniscal tears situated within the inner, avascular region do not heal. We investigated the potential effect of insulin-like growth factor-I (IGF-I) in promoting regeneration of meniscal tissue in the inner, middle and outer zones of the meniscus. Sheep menisci were harvested and monolayer cell cultures prepared. Various concentrations of IGF-I were used in the presen...

Tissue-engineered Meniscal Replacement in Dogs Using Small Intestinal Submucosa: Phase 2, Addition of Autogenous Fibrin Clot and Biomechanical Protection

Introduction Meniscal injuries in human and veterinary patients result in a significant loss of function, and those occurring in the avascular zone of the meniscus will not heal spontaneously. Although most meniscal injuries are currently treated by partial meniscectomy, most research is now focused on methods for producing functional tissue in avascular meniscal defects. We have previously dem...

Gene Therapy for Cartilage Repair

The concept of using gene transfer strategies for cartilage repair originates from the idea of transferring genes encoding therapeutic factors into the repair tissue, resulting in a temporarily and spatially defined delivery of therapeutic molecules to sites of cartilage damage. This review focuses on the potential benefits of using gene therapy approaches for the repair of articular cartilage ...

Evaluation of small intestinal submucosa grafts for meniscal regeneration in a clinically relevant posterior meniscectomy model in dogs.

Large meniscal defects are a common problem for which treatment options are limited. Successful meniscal regeneration has been achieved by using grafts of small intestinal submucosa in posterior, vascular meniscal defects in a dog model. This study investigates the long-term effects of a tibial tunnel fixation technique and a clinically based meniscectomy defect on meniscal regeneration using t...

Transplantation of Achilles Tendon Treated With Bone Morphogenetic Protein 7 Promotes Meniscus Regeneration in a Rat Model of Massive Meniscal Defect

OBJECTIVE This study was undertaken to examine whether bone morphogenetic protein 7 (BMP-7) induces ectopic cartilage formation in the rat tendon, and whether transplantation of tendon treated with BMP-7 promotes meniscal regeneration. Additionally, we analyzed the relative contributions of host and donor cells on the healing process after tendon transplantation in a rat model. METHODS BMP-7 ...
